The legacy PC app supports USB microphones. Make sure you select the microphone under Tools > Options > Audio > Recording device. 

PTT can triggered using HID. So you can use a keyboard, and map one of the keys to the Zello PTT button, or any other accessory that uses HID such as a foot pedal.
